To do this, you will have to use something other than just HTML or javascript. You can use activeX or even an applet. There are a few of these scattered all over the internet. Some examples:

Hello -

I would very much like to find a way to drag and drop onto a web browser from the OS level.  The dragged files would be uploaded to a server, so probably this can't work without sometype of script signing or something.  Is anything like this possible?

I've found that some d&d js scripts i've found will recognize dragging from the desktop, but the contents are null.

I'm assuming all I can do is write an activex control.  Is that true?
